SUMMARY:

Leishmaniasis is an infectious, non-contagious disease whose etiological agent is protozoa of the genus Leishmania. It is a disease with a clinical spectrum. Although it is endemic in various regions of the world, and considered a public health problem in many countries, it is observed that the existing pharmacological arsenal for the treatment of leishmaniasis is limited by a number of factors, such as high toxicity, a variable variable, parasite-acquired resistance that leads to an increase in treatment failure and longer treatment regimens. That said, it is observed that the existing pharmacotherapy is often unsatisfactory, which makes the planning and development of new drugs an essential condition. Thus, there is a need to find new prototypes for leishmanicidal drug candidates. At this juncture, the antineoplastic and antipastitis activity of a class of compounds known as derivatives of purpurogaline, a natural benzotropolone, which can be easily installed synthetically in the laboratory, has been applied in the literature. Thus, 8 derivatives of purpurogaline were tested in order to investigate the leishmanicidal activity against Leishmania chagasi. Some of the derivatives tested did not demonstrate any harmful effect on the host cell, with the exception of derivatives IAS 2 and IAS 5. In the assessment of direct activity on L. chagasi promastigotes, derivatives IAS 1 to IAS 6 maximum effect (MS) greater than 70% and IC50 less than 15 µM. Such results indicate that IAS 1 to 6 derivatives, significant leishmanicidal activity, in vitro and, therefore, can be considered candidates to advance in the of studies of new leishmanicidal agents.
