SUMMARY:

Chikungunya virus (CHIKV) is an Alphavirus belonging to the Togaviridae family, transmitted mainly by the bite of infected Aedes aegypti or Aedes albopictus mosquitoes. In the acute phase, it presents with symptoms common to other arboviral diseases, such as fever, myalgia, arthralgia, pruritus, and headache. In the chronic phase, symptoms such as persistent polyarthritis and/or polyarthralgia are reported, and neurological complications may also arise, such as encephalitis, meningoencephalitis, Guillain-Barré syndrome, and congenital infections. Despite the clinical relevance of CHIKV, there are still no approved antiviral drugs for its treatment. Previous studies have shown that peptidomimetics containing the Arg-Gly-Gly-Leu sequence exhibit activity against the CHIKV nsP2 protein. nsP2 is a viral protease essential for replication, being responsible, through its C-terminal autoprolytic domain, for the cleavage of the P1234 polyprotein, releasing the non-structural proteins necessary for the formation of the viral replication complex. This processing is essential for viral RNA synthesis and, consequently, for the production of new virions. In this project, new peptidomimetics were designed, considering the Y↓GGAR sequence, identical to the nsP2 substrate, and modifications were made in the N-terminal region based on precursors previously synthesized and tested by our group, with the aim of increasing ligand affinity and thus improving their biological potential.
