Ranitidine is a histamine H2-receptor antagonist employed in the treatment of gastroduodenal ulcer and related diseases

such as gastro-oesophageal reflux and Zollinger-Ellison Syndrome. Its main function is to inhibit overproduction of
gastric acid due to its effect on gastric acid secretion and thus reduce the pain. Listed by the world health organization
(WHO) as one of the essential medicines for use in the basic health system, ranitidine has a global market and is
commercially available under the brand Zantac and others.
Several analytical methods have been reported for ranitidine determination, including electrochemical
spectophotometric, chromatography, fluorescence and chemiluminescence. Although these methods are suitable for
ranitidine determination in different samples, they present major drawbacks including high cost and reagent consuption
, laborious operation, complex equipments and trained personnel. Thus, methods that overcome these drawbacks are
required and demanded.
Paper-based analytical devices (PADs) have emerged as a powerful tool due to its characteristics such as low
cost, portability, low reagent consuption and ease of use. Since the pioneering research developed by the Whiteside’s
group in 2007, applications in several fields such as forensics, drug analysis and medicine have been reported.
PADs with colorimetric methods have gained attention due to the ease of handling, simplicity, low cost equipaments
and signal acquisition with the use of scanners, smartphones or digital cameras. Besides, the color analysis generated
can be performed with the aid of software or smartphone applications, which makes the use of these devices essential
in remote regions or in developing countries. Moreover, the versatility of colorimetric detection based on redox reactions,
pH changes, nanoparticles or complex formation allows the use of these methods in several fields of analysis.
Herein, a spot test paper-based analytical device for the determination of ranitidine in pharmaceutical samples is
presented. The methodology is based on the reaction between ranitidine and the colorimetric probe 4-Chloro-7-
nitrobenzofurazan (NBD-Cl), generating a color proportional to the concentration of ranitidine. This methodology proved
to be suitable for application in pharmaceutical samples and is a simple, cheap, and fast alternative for the determination
of ranitidine.
