SUMMARY:

In recent years, there has been a significant increase in research that seeks to develop alternative technologies, through the use of raw materials without commercial value, among which agro-industrial residues, such as sugarcane bagasse (BCA), fiber coconut (FC), pineapple crown (CA) and wheat bran (FT), discarded, in most cases, inappropriately, causing environmental impacts. Among the most used enzymes worldwide, hydrolases stand out, where proteases and glycosities are classified as extremely important enzymes for the maintenance of organisms and biotechnological applications, and may come from animal, vegetable and microbial sources. In this work, the partial isolation of an endoglucanase and a protease was carried out through the solid state cultivation of the filamentous fungus species Pleurotus djamor PLO13 in wheat bran residue. For this, four residues from agroindustry, BCA, FC, CA and FT were initially used as support for the growth of the fungus in order to observe the best mycelial development, as the FT showed greater growth efficiency, after 120h of cultivation, when compared to the others. Then, enzymatic assays were carried out to analyze the presence of endoglucanase and protease in the extract, being verified the presence of both. Soon after, saline and organic fractions were carried out in order to concentrate and, consequently, start the enzyme isolation process. It was observed that organic fractionation with ethanol was more effective for these hydrolases, since they were concentrated in the 100% fraction, showing high enzymatic activity with recovery greater than 100%. From the analysis of the SDS-PAGE gel, it was possible to verify the efficiency of the experiment, since the concentration and separation of proteins present in the sample was observed throughout the stages. When performing coagulation tests with pasteurized skim and whole bovine milk, it was observed that the enzyme extract has at least one protease with high coagulant activity, highly efficient and little dependent on calcium. It was also observed that this new coagulating agent presented enzymatic activity to LEU and PHE substrates and was inhibited by EDTA and DTT, the enzyme degraded the gelatin present in the zymography gel, suggesting that it is a metalloprotease with optimal activity of 50 ºC ., such results show that the enzymes have a high potential for use in industrial sectors, and can be used, respectively, in the production of second-generation ethanol and dairy industries, mainly in the production of cheese.
