SUMMARY:

Chagas disease and Human African Trypanosomiasis are caused by Trypanosoma cruzi and T. brucei, respectively. Cruzain (CRZ) from T. cruzi and cathepsin L-like from T. brucei (TbrCATL) are cysteine proteases considered promising targets for the development of new compounds with trypanocidal activity, since they play vital roles in these parasites. Coumarin derivatives have been reported as promising trypanocidal agents. In this sense, this study aims to synthesize rationally designed coumarin-derived compounds targeting such cysteine proteases from T. cruzi and T. brucei protozoa. Therefore, to discover potential inhibitors of such targets, the rational design of compounds based on molecular docking studies was applied. Subsequently, the biological evaluation of the derivatives and in silico studies of the most promising compounds will be carried out. Finally, it is expected, at the end of this work, to produce potentially active molecules against the T. cruzi and T. brucei parasites, through enzymatic inhibition.
