SUMMARY:

Brazilian red propolis is a complex mixture, waxy in nature, made from substances collected from plant parts, buds and exudates that are mixed with the saliva of Apis mellifera bees. Used since ancient times, it has important antimicrobial and anti-inflammatory activity. This study aimed to produce dental floss with antimicrobial activity against cariogenic bacteria. An ethanolic extract of Brazilian red propolis (PVB) was obtained and analyzes were then carried out to track down the phytochemical profile of the propolis used: 1-high performance liquid chromatography (HPLC), 2- quantifying of flavonoids and total phenols and 3-antioxidant activity (DPPH). After the first phase of obtaining and characterizing the PVB extract, polytetrafluoroethylene (PTFE) dental floss impregnated with PVB at concentrations of 500, 1000, 2000 and 3000 mg/mL were produced. Then, the dental floss were analyzed using infrared spectrometry (FTIR) and the antioxidant activity tested (DPPH). The surface morphology of the dental floss was evaluated using scanning electron microscopy (SEM). The antimicrobial activity of dental floss impregnated with PVB at experimental concentrations will be tested against strains of Streptococcus mutans CCT 3440, Lactobacillus acidophilus ATCC 4356, Candida albicans ATCC 36801 and 36802 using the disk diffusion method (DDM). The propolis extract will be analyzed to determine the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC). The chromatogram showed the presence of the isoflavonoids daidzein, liquiritigenin, pinobanksin, isoliquiritigenin, formononetin, pinocembrin and biochanin in the 500µg/mL PVB extract. With higher amounts of liquiritigenin (6.20mg/mL), isoliquiritigenin (6.19mg/mL) and formononetin (6.90mg/mL). The dental floss showed antioxidant capacity. SEM images show that the untreated sample had a smooth surface. After treatment with potassium permanganate and nitric acid, the surface changes, becoming more porous and with some cracks. Dental floss treated and soaked in PVB extract seems to have been covered with a substance that gave it a more uniform surface. The antimicrobial activity still will be tested; however, it is expected that the floss will have good capacity anticariogenic. With the methodology developed so far, it was possible to produce a PTFE dental floss coated with PVB at the concentrations proposed in this study. which have antioxidant activity.
