SUMMARY:

Red propolis is a resinous mixture of substances collected from certain species of plants by bees of the Apis mellifera species, with a complex composition and has been studied for various therapeutic purposes over the years. The hydroalcoholic extract of red propolis has some drawbacks with high alcohol content and low solubility in water. To solve these problems, this scientific work presents red propolis granules in the solid state with advantages over hydroalcoholic extracts of red propolis from Alagoas, as they are free of alcohol, have good solubility in aqueous GIT fluids, and are presented in the form pharmaceutical in stable solid state, with good loading capacity of bioactive compounds for oral use and consequently reduces the number of doses used during the day, thus improving therapeutic adherence. The present work developed PVA granules from the 70ºGL hydroalcoholic extract acquired by the company Beeva. The Beeva extract was evaporated and concentrated, and then used in the production of granules with the addition of microcrystalline cellulose. The concentrated ethanolic extract, microcrystalline cellulose and PVA granulate were subjected to chemical and physicochemical characterization tests by phytochemical screening tests total phenols, total flavonoids, antioxidant activity tests, UV-Vis tests and UPLC-DAD, test SEM, thermal analysis (TG; DSC), infrared analysis and dissolution tests. The results of characterizing the crude extract and phenolic compounds of red propolis such as isoflavonoids using the comparative technique of injections of analytical standards by UPLC-DAD showed that the granulate presents similar concentrations to the extract without loss of marker concentration. A physicochemically stable composition of the PVA-cellulose granulate was developed when observed by the TG technique. Scanning Electron Microscopy analysis demonstrated that the granulate appeared as an amorphous powder. Fourier Transform Infrared (FTIR) analysis showed O-H stretching of hydroxyl (3300 cm-1), C-H stretching of CH2, and CH3 at 2900 cm-1, characteristic aromatic ring stretching (1600, 1500 and 1450 cm-1). 1) and C-O-C stretching (1050 cm-1) of the ether moiety present in the flavonoids that appear more intensely in the PVA Extract than in the PVA granulate. The granulate presented a dissolution percentage of 80% in the 0.1% LSS medium and 92% in the 1% Tween medium. The PVA Extract and PVA-Cellulose granules showed antioxidant activity in the concentration range between 5 and 75 µg/mL with an IC50 of 10 µg/mL for the PVA Extract and 5 µg/mL for the PVA granules. PVA granules can be used as a new pharmaceutical form of Alagoas red propolis extracts.
