SUMMARY:

The medicinal plant Uncaria tomentosa has been widely used in traditional medicine in Peru, where its decoctions, prepared from its bark, are used to treat various diseases, in particular as a potent anti-inflammatory agent. In vitro studies indicate that the anti-inflammatory activity of the U. tomentosa extract is related to the prevention of the production of the transcription factor NF-kβ (nuclear factor kappa B) at the molecular level thus inhibiting the transcription of genes associated with the inflammatory process. The work developed and characterized spray-dryer microcapsules of Uncaria tomentosa extract. The husks of Uncaria tomentosa were acquired by the company Kampo de Ervas® and subjected to extraction by infusion and extraction by maceration using hydro-ethanolic solution (70ºGL and 92ºGL). The hydro-ethanolic solutions were used for partitioning and enrichment following the march of alkaloids and subsequent testing for antibacterial activity. The 92 ° GL ethanolic extract was used to prepare a spray-dryer dry extract composition. The infusion extracts, 92ºGL ethanolic extracts and spray-dryer dry extract from Uncaria tomentosa were subjected to characterization assays by assays of total phenols, total flavonoids, antioxidant activity assays, UV-Vis and CLAE-DAD-UV assays, SEM test, thermal analysis tests, infrared test and dissolution tests. The results obtained in the study of obtaining and characterizing the crude extracts and microcapsules of Uncaria tomentosa reveal that the ethanolic extract and microcapsules of Uncaria tomentosa showed the presence of alkaloids and phenolic compounds using data from high-performance liquid chromatography, chemical tests and phytochemical screnning. The hydro-ethanolic extract and dichloromethane fractions of Uncaria tomentosa showed antibacterial activity probably due to the synergistic effect between alkaloids and phenolic compounds. The ethanolic extracts and microcapsules of Uncaria tomentosa demonstrated antioxidant activity comparable to studies in the scientific literature. A physically-chemically stable composition of spray-dryer microcapsules from Uncaria tomentosa with micrometric size when analyzed by Scanning Electron Microscopy, FTIR and Thermal Analysis. The spray-dryer microcapsules of Uncaria tomentosa proved to be poorly soluble in water, which were optimized during in vitro dissolution tests for better absorption of the active constituents by gastrointestinal fluids by the addition of pharmacotechnical adjuvants and consequently resulting in solid phytotherapeutic compositions with guaranteed release efficacy.
