SUMMARY:

Malaria is considered one of the most serious parasitic diseases of this class, being often fatal, with a high rate of transmission in countries with tropical climates. In 2018 alone, it is estimated that there were 228 million cases and 405,000 deaths from the disease. Such alarming numbers point to a deficiency in antimalarial treatment triggered by the resistance of the parasites that cause malaria, mainly by plasmodium falciparum against chloroquine, making it necessary to develop new antimalarial agents active against these resistant strains. Therefore, this work aimed to synthesize aminoquinoline compounds that already showed efficient antiparasitic activities (in vitro and in vivo) and modify them by hydrogenation in heterogeneous catalysis in an attempt to increase their antimalarial activity. The compound that underwent modification was N-(4-(diethylamino)but-2-ynyl)-7-chloro-quinolin-4-amine (DAQ), synthesized in 40% yield from N-(prop- 2-ynyl)-7-chloro-quinolin-4-amine (PCQ) in 92% yield. The modification occurred in its aliphatic chain, where there is a triple bond, having as a product of this modification a mixture of olefinic molecules, as well as molecules with the aliphatic chain fully saturated by the addition of hydrogens. All compounds, as well as the aforementioned mixture, were characterized by Nuclear Magnetic Resonance Spectroscopy for the Hydrogen nucleus. The next step of this work is to obtain the compounds, originating from the modification, so that they are isolated for further spectroscopic characterization and biological tests, which will guarantee their quality and their potential value as antimalarial drugs.
