Durante os processos de formação e progressão tumoral há o envolvimento da ação de células que criam
um microambiente favorável para o desenvolvimento do câncer. Dentre os mecanismos que atuam nesses
eventos, encontram-se alterações de ritmo circadiano e invasão perineural. Ambos os eventos foram
descritos, separadamente, como atuantes na agressividade tumoral. É importante identificar eventos relacionados
à alteração do ritmo circadiano que são associados ao câncer que possam impactar diretamente
na invasão deste tecido no sistema nervoso periférico. Nesse contexto, o objetivo do presente
estudo foi avaliar a invasão perineural em câncer de cabeça e pescoço, associando a genes relacionados
ao sistema nervoso periférico e ao ritmo circadiano. Os dados demonstram que as vias de “ritmo circadiano”
e “orientação axônica” apresentaram-se desreguladas em carcinoma epidermoide de cabeça e
pescoço com invasão perineural positiva. As análises da expressão diferencial identificaram que os genes
associados à resposta das células de Schwann em nervos lesados que são regulados positivamente
ativam a via do “ritmo circadiano”. Foi identificado correlação negativa entre a expressão e a metilação
dos dos genes PER3 e RORC, quando analisados separadamente em ambos grupos tumorais. As correlações
positivas entre as expressões de PER2 e RORC, bem como de RORC com os genes MPZ, S100A8
e NUF2, foram identificadas apenas nos grupos de pacientes que não apresentaram invasão perineural.
Os genes do ritmo circadiano estão desregulados nos tecidos neoplásicos, estando os oncogenes com a
expressão aumentada e os supressores de tumor com a expressão diminuída. O aumento da expressão
da proteína mTOR foi identificada nos pacientes que apresentam invasão perineural. Os dados sugerem
que a desdiferenciação e proliferação das células de Schwann podem estarem associadas à desregulação
de genes do ritmo circadiano; consequentemente, as células de Schwann produzem diferentes fatores
que participarão de diversos processos de progressão tumoral e invasão dos tecidos adjacentes, incluindo
ativação da via mTOR. Esses processos também podem estar envolvidos na invasão tumoral no tecido
perineural no câncer de cabeça e pescoço.

During the processes of tumor formation and progression there is the involvement of the action of cells
that create a favorable microenvironment for cancer development. Among the mechanisms involved in
these events are circadian rhythm alterations and perineural invasion. Both events have been described,
separately, as active in tumor aggressiveness. It is important to identify events related to circadian
rhythm alterations that are associated with cancer that may directly impact the invasion of this tissue
into the peripheral nervous system. The aim of this study was to evaluate perineural invasion in head
and neck cancer, associating it with genes related to the peripheral nervous system and circadian rhythm.
The data showed that the circadian rhythm and axon guidance pathways were deregulated in head and
neck squamous cell carcinoma with positive perineural invasion. Differential expression analyses identified
that genes associated with Schwann cell response in injured nerves that are positively regulated
activate the circadian rhythm pathway. Negative correlation was identified between expression and
methylation of PER3 and RORC genes when analyzed separately in both tumor groups. Positive correlations
between expression of PER2 and RORC, as well as of RORC with MPZ, S100A8 and NUF2
genes were identified only in the groups of patients who did not present perineural invasion. Circadian
rhythm genes are dysregulated in neoplastic tissues, with increased expression of oncogenes and decreased
expression of tumor suppressors. Increased expression of mTOR protein was identified in patients
presenting with perineural invasion. The data suggest that dedifferentiation and proliferation of
Schwann cells may be associated with downregulation of circadian rhythm genes; consequently,
Schwann cells produce different factors that participate in various processes of tumor progression and
invasion of adjacent tissues, including activation of the mTOR pathway. These processes may also be
involved in tumor invasion into the perineural tissue in head and neck cancer.

O vírus Chikungunya (CHIKV) é um arbovírus transmitido por mosquitos do gênero Aedes sp., capaz de causar a febre Chikungunya (CHIKF) em sua fase aguda e evoluir para uma Síndrome Reumática Crônica (SRC). Nesta infecção, tem-se o acometimento das articulações sinoviais que apresentam em sua composição os sinoviócitos semelhantes a fibroblastos (FLS). Os FLS, em patologias semelhantes à SRC como a artrite reumatóide (AR), apresentam um perfil agressivo capaz de causar danos teciduais severos no processo inflamatório, processo este que pode ocorrer devido a alterações em propriedades biomecânicas celulares. Diante disso, o presente trabalho objetivou identificar alterações biomecânicas, bioquímicas e moleculares em sinoviócitos semelhantes a fibroblastos humanos (HFLS) após a infecção pelo CHIKV in vitro. Para isso, foi realizada a determinação da MOI por MTT e a confirmação da infecção viral por citometria de fluxo intracelular, RT-PCR e infecção de células Vero E6 com sobrenadante de cultivo de HFLS. Foram avaliadas: alterações morfológicas e o módulo elástico de Young através de microscopia de força atômica (MFA); alterações no citoesqueleto por ensaio de fluorescência com marcação para F-actina; alterações bioquímicas através de espectroscopia Raman; alterações na expressão gênica por RT-qPCR; e quantificação quimiocinas no sobrenadante de cultivo através da metodologia de CBA. Os resultados indicaram que os HFLS foram infectados in vitro, observando-se uma porcentagem de 46,8% de células positivas para o CHIKV após 48h através de citometria de fluxo intracelular, além da identificação do genoma viral no sobrenadante por RT-PCR. A presença de unidades formadoras de placas (PFUs) na monocamada de células Vero E6 infectadas indicou a presença de partículas virais viáveis no sobrenadante dos HFLS. A análise por MFA evidenciou alterações significativas na morfologia celular e as análises de nanoindentação detectaram um aumento de 107,46% no módulo elástico de Young, sugerindo que a infecção pelo CHIKV pode levar a alterações no citoesqueleto. Análise do citoesqueleto por fluorescência indicaram rompimento dos filamentos de F-actina após 12h de infecção. A espectroscopia Raman e a subsequente análise de PCA indicaram uma variância total de 65% entre os HFLS não infectados e os HFLS infectados pelo CHIKV. Além disso, os principais picos alterados estão relacionados a alterações em nucleotídeos, amidas primárias e terciárias, folhas β, lipídeos e colágeno, sendo eles os picos 795, 977, 1031, 1223, 1290, 1444, 1495, 1635, 1660 e 1709 cm-1. A análise de expressão gênica indicou um aumento na expressão dos genes MMP1, VEGFA e UGDH. Além disso, um aumento significativo de IP-10 e uma diminuição de MCP-1 foram observadas no sobrenadante 48h após a infecção viral. Em resumo, os resultados apresentados no presente trabalho auxiliam na elucidação das alterações celulares e moleculares envolvidas na interação CHIKV-HFLS, contribuindo para o desenvolvimento de novas estratégias terapêuticas para esta arbovirose.

Chikungunya virus (CHIKV) is an arbovirus transmitted by Aedes sp. mosquitoes that causes Chikungunya fever (CHIKF) and could be progress to Rheumatic Chronic Syndrome (RSC). The synovial joints are affected during this infection, and fibroblast- like synoviocytes (FLS) are the major cell type in the composition of synovial tissue. FLS in pathologies such as rheumatoid arthritis (RA), assumes an aggressive profile, causing severe tissue damage in the inflammatory process. This damage can occur due to changes in cellular biomechanical properties. Thus, the aim of this work was to understand biomechanical, biochemical and molecular changes in human fibroblast-like synoviocytes (HFLS) after CHIKV infection in vitro. Determination of the MOI was performed by MTT assay and viral infection was confirmed by flow cytometry, RT-PCR and infection of Vero E6 cells with HFLS supernatant. The following were evaluated: morphological changes and Young`s modulus by atomic force microscopy (AFM); cytoskeleton changes by fluorescence analysis of F-actin labeling; biochemical changes were determined by Raman spectroscopy; relative gene expression analysis by RT-qPCR. Also, the quantification of chemokines was measured in the supernatant using the CBA methodology. As result, HFLS were infected in vitro since 46.8% of the cells were CHIKV-positive after 48h of infection in the intracellular flow cytometry analysis. In addition, viral RNA in the cell supernatant was detected by RT-PCR. The detection of plaque-forming units (PFUs) in the monolayer of infected Vero E6 cells indicated the presence of viable viral particles in the HFLS supernatant. AFM analysis shows significant changes in cell morphology and nanoindentation analysis detected a 107.46% increase in Young's elastic modulus, suggesting alterations in the cytoskeleton. Fluorescence cytoskeleton staining indicated disruption of F-actin filaments after 12h of infection. Raman spectroscopy and PCA analysis show a total variance of 65% between uninfected and CHIKV-infected HFLS. In addition, the major alterations detected were related to nucleotides, amide I, amide III, β-sheets, lipids and collagen, with signatures in 795, 977, 1031, 1223, 1290, 1444, 1495, 1635, 1660 and 1709 cm-1. Relative gene expression shows an increase in the expression of MMP1, VEGFA and UGDH. Also, an increase in the IP-10 and a reduction in MCP-1 chemokine levels were detected in the supernatant 48 after viral infection. In summary, the results presented in this work help elucidate the cellular and molecular changes involved in the CHIKV-HFLS interaction, thus contributing to developing new therapeutic strategies for this arbovirus.

O melanoma é considerado um dos tipos neoplásicos mais agressivos por apresentar rápida evolução para metástase, embora vários fatores ambientais e genéticos já tenham sido descridos podendo influenciar no desenvolvimento e agravo desse tipo tumoral a desregulação no sistema de temporização circadiana permanece recebendo pouca atenção. A melatonina é o principal hormônio controlador dos genes do relógio biológico, sabe-se que uma eventual mudança na regulação desses genes pode contribuir para o desenvolvimento de várias doenças inclusive o câncer. Neste estudo, foram realizadas análises de bioinformática com dados de tecido de melanoma e câncer de pele do tipo não melanoma a fim de avaliar quais genes relacionados ao relógio biológico podem contribuir para a progressão desse tipo tumoral. Além disso, ensaios de cultura celular foram realizados utilizando a melatonina como tratamento em linhagens celulares de carcinoma cutâneo não melanoma (A431) e no tipo melanoma (B16) visando observar alterações nessas células neoplásicas. Modificações nas expressões gênicas associadas aos genes do relógio, proliferação, migração, invasão e apoptose celular nas células A432 e B16 após o tratamento com a melatonina foram avaliadas. Em resumo, os genes relacionados ao relógio biológico apresentaram expressões alteradas no câncer de pele do tipo melanoma e esses genes foram associados com uma baixa sobrevida de acordo com a análise de bioinformática. Em cultura celular, a administração de melatonina aumentou a expressão dos genes PER2, CRY1 e CRY2 e diminuiu os níveis de expressão do gene CLOCK. Os genes PER2, CRY1 E CRY2 são considerados genes supressores tumorais que podem controlar eventos de importância no contexto tumoral como a proliferação celular, apoptose e controle do sistema de reparo e dano ao DNA. A administração de melatonina mostrou ser capaz de induzir alterações comportamentais nas células do melanoma e influenciar as expressões dos genes MKI67, HIF-1A, VEGFA, COL1A1, CDH1, CASP3, BAX, BCL2, MMP9, PER2, CRY1, CRY2 E CLOCK. A principal causa de mortalidade pelo melanoma é a disseminação metastática e as complicações subsequentes, apesar dos avanços na cirurgia e em outros métodos de tratamento, a taxa de sobrevida de um paciente com melanoma metastático permanece inalterada, apenas 4 anos, tendo em vista os dados aqui relatados, espera-se contribuir com a pesquisa terapêutica visando o tratamento de indivíduos que apresentem essa condição.

Melanoma is considered one of the most aggressive neoplastic types because it presents a rapid progression to metastasis, although several environmental and genetic factors have already been described that may influence the development and aggravate this tumor type, the deregulation in the circadian timing system remaining unattractive. Melatonin is the main hormone controlling the genes of the biological clock, it is known that an eventual change in the regulation of these genes can contribute to the development of several diseases including cancer. In this study, bioinformatics analyzes were performed with tissue data from melanoma and non-melanoma skin cancer in order to assess which genes related to the biological clock may contribute to the progression of this tumor type. In addition, cell culture assays were carried out using melatonin as a treatment in non-melanoma cutaneous carcinoma cell lines (A431) and melanoma-type (B16) observing changes in these neoplastic cells. Changes in gene expression associated with clock, anticipation, migration, invasion and cell apoptosis genes in A432 and B16 cells after melatonin treatment were evaluated. In summary, genes related to the biological clock showed altered expression in melanoma skin cancer and these genes were associated with poor survival, according to the bioinformatics analysis. In cell culture, melatonin administration increased the expression of the PER2, CRY1 and CRY2 genes and experienced CLOCK gene expression levels. The PER2, CRY1 and CRY2 genes are considered tumor suppressor genes that can control events of importance in the tumor context, such as cell anticipation, apoptosis and control of the DNA repair and damage system. Melatonin administration has been shown to be able to induce behavioral changes in melanoma cells and influence the expression of genes MKI67, HIF-1A, VEGFA, COL1A1, CDH1, CASP3, BAX, BCL2, MMP9, PER2, CRY1, CRY2 AND CLOCK. The main cause of mortality from melanoma is metastatic dissemination and subsequent complications, despite advances in surgery and other methods of treatment, the survival rate of a patient with metastatic melanoma remains unchanged, only 4 years, in view of the data reported here, it is expected to contribute to therapeutic research by guiding the treatment of individuals who have this condition.

O uso de macromoléculas obtidas de tecidos vegetais apresenta grande potencial econômico e farmacológico e isso se dá em decorrência da presença dos compostos bioativos. Dentre estes, lectinas veem sendo intensamente exploradas devido as diversas ações biotecnológicas que podem ter, destacando-se as atividades antifúngica, anticoagulante e antitumoral. Desse modo, o presente trabalho teve por objetivo purificar, caracterizar e avaliar a atividade antitumoral, antimicrobiana e sobre parâmetros hemostáticos das lectinas de Genipa americana L. (jenipapo), Rhizophora mangle L. (mangue vermelho) e da própolis vermelha de Alagoas. As lectinas foram isoladas inicialmente através de extração em Tris-Hcl 50mM pH 8.0, para sementes e cascas de G. americana), NaCl 0,15M, para sementes de R. mangle, e NaCl 0,15M com com 30 % de etanol para a própolis vermelha. Em seguida, foram feitos fracionamentos salinos com sulfato de amônio e as frações que apresentaram melhor atividade foram submetidas à cromatografia de exclusão molecular (Sephacryl S-100), para isolamento das lectinas de casca de G. americana (GaBL); cromatografia de exclusão molecular (Sephacryl S-100) seguida de uma trocadora catiônica (DEAE Sepharose) para a lectina de sementes de G. americana (GaSL); ou cromatografia em coluna de quitina para isolar as lectinas de R. mangle e da própolis vermelha. Através de um único passo cromatográfico foi isolada uma lectina (GaBL) e por meio de SDS-PAGE (10%) foi observado que GaBL apresenta uma massa molecular aproximada de 242,5 kDa. No teste citotóxico GaBL não se mostrou tóxica para linhagens celulares de fibroblastos 3T3 em concentrações abaixo de 50μg/mL. O efeito da lectina na coagulação sanguínea foi avaliado através do tempo de tromboplastina parcial ativada (TTPA), tempo de protrombina (TP). GaBL apresentou atividade anticoagulante somente na via intrínseca, onde o TTPA apresentou-se prolongado. A atividade antitumoral avaliada utilizou as linhagens celulares de câncer de pele humano (A431), melanoma (B16) e carcinoma de células escamosas da língua (SCC9). A proliferação celular e a migração celular foram diminuidas em todas as linhagens avaliadas em contato com 10 µg/ml de GaBL diminuiu a invasão de células SCC9. A apoptose foi maior nas células B16 e SCC9 tratadas com lectina. A regulação positiva de GaBL de E-caderina e supressão de Col1A1 em todas as cepas testadas indicou um menor desenvolvimento de câncer. (A lectina de sementes de G. americana, GaSL, teve sua atividade hemaglutinante (AH) inibida fortemente por ramnose, mas não foi afetada por íons divalentes nem por EDTA. GaSL é classificada como uma lectina termoestável, com uma melhor atividade a uma temperatura abaixo de 80 ° C e na faixa de pH 5,0 a 6,0. Frente as atividade antifúngicas, GaSL quando testada com os fungos Candida albicans, Staphylococcus aureus e Cryptococcus neoformans, mostrou-se inibir todas as cepas fungicas em concentrações abaixo de 12,5 μg/mL. A lectina de sementes de R. mangle é também termoestável, mas, diferente das demais, foi inibida mais intensamente por arabinose e caseína, teve melhor atividade em pH 8,0 e 9,0 e teve sua AH reduzida em presença de íons Mn2+, Mg2+ e Ca2+, não alterada por EDTA e estimulada em presença de Zn2+. A lectina da própolis também foi inibida fortemente por caseína, é termoestável, apresentou melhor AH na faixa de pH 5,0 a 6,0 e foi reduzida em presença de Mn2+, Mg2+, levemente estimulada por Zn2+ e não afetada por EDTA e Ca2+. Frente a atividade antifúngica PVAL se mostrou uma promissora ferramenta biotecnológica, quando testada com os fungos Candida albicans, Staphylococcus aureus e Cryptococcus neoformans. PVAL nas concentrações 12,5 μg/mL foi capaz de inibir fortemente Candida albicans e Staphylococcus aureus e 25 μg/mL Cryptococcus neoformans. O efeito de PVAL na coagulação sanguínea foi avaliado através do tempo de tromboplastina parcial ativada (TTPA), tempo de protrombina (TP). Onde PVLA promoveu expressivo prolongamento de TTPA, enquanto que para TP PVAL promoveu forte inibição. Diante Dos dados expostos, podemos destacar que GaBL tem elevado potencial antitumoral contra câncer do trato aero digestivo superior e possue notória atividade homeostática. GaSL e PVAL possui notável atividade antifúngica podendo ser uma promissora ferramenta biotecnológica, tendo esta ultima um importante papel nos processos homeostáticos da coagulação, sendo um forte prolongador da TTPA e inibidor de TP. E por fim destacamos que todas as lectinas estudadas apresentam características estruturais que favorecem o estudo biotecnológico envolvendo a saúde humana.

The use of macromolecules obtained from plant tissues has great economic and pharmacological potential and this is due to the presence of bioactive compounds. Among these, lectins have been intensely explored due to the various biotechnological actions they may have, especially the antifungal, anticoagulant and antitumor activities. Thus, the present work aimed to purify, characterize and evaluate the antitumor, antimicrobial and hemostatic activity of lectins from Genipa americana L. (jenipapo), Rhizophora mangle L. (red mangrove) and red propolis from Alagoas. Lectins were isolated initially by extraction in 50mM Tris-Hcl pH 8.0 for G. americana seeds and bark, 0.15M NaCl for R. mangle seeds and 0.15M NaCl with 30% ethanol for red propolis. Then, salt fractions were made with ammonium sulfate and the fractions that showed better activity were submitted to molecular exclusion chromatography (Sephacryl S-100), for isolation of lectins from G. americana (GaBL); molecular exclusion chromatography (Sephacryl S-100) followed by a cation exchanger (DEAE Sepharose) for the lectin from G. americana seeds (GaSL); or chitin column chromatography to isolate lectins from R. mangle and red propolis. Through a single chromatographic step a lectin (GaBL) was isolated and by SDS-PAGE (10%) it was observed that GaBL has an approximate molecular mass of 242.5 kDa. The effect of lectin on blood clotting was evaluated through activated partial thromboplastin time (APTT), prothrombin time (PT). GaBL showed anticoagulant activity only in the intrinsic pathway, where the APTT was prolonged.
In the cytotoxic test, GaBL was not toxic to 3T3 fibroblast cell lines at concentrations below 50μg/mL. The antitumor activity evaluated used human skin cancer (A431), melanoma (B16) and tongue squamous cell carcinoma (SCC9) cell lines. Cell proliferation and cell migration were decreased in all cell lines evaluated in contact with 10 µg/ml of GaBL decreased invasion of SCC9 cells. Apoptosis was higher in lectin treated B16 and SCC9 cells. GaBL's positive regulation of E-cadherin and suppression of Col1A1 in all strains tested indicated less cancer development. The lectin from G. americana seeds, GaSL, had its hemagglutinating activity (AH) strongly inhibited by rhamnose, but was not affected by divalent ions or EDTA. GaSL is classified as a thermostable lectin, with best activity at a temperature below 80°C and in the pH range 5.0 to 6.0. In the face of antifungal activity, GaSL, when tested with the fungi Candida albicans, Staphylococcus aureus and Cryptococcus neoformans, was shown to inhibit all fungal strains at concentrations below 12.5 μg/mL. The lectin from R. mangle seeds is also thermostable, but unlike the others, it was inhibited more strongly by arabinose and casein, had better activity at pH 8.0 and 9.0 and had its AH reduced in the presence of Mn2+, Mg2+ and Ca2+ ions, not altered by EDTA and stimulated in the presence of Zn2+. The lectin from propolis was also strongly inhibited by casein, is thermostable, showed better AH in the pH 5.0 to 6.0 range and was reduced in the presence of Mn2+, Mg2+, slightly stimulated by Zn2+ and not affected by EDTA and Ca2+. In view of the antifungal activity PVAL proved to be a promising biotechnological tool, when tested with the fungi Candida albicans, Staphylococcus aureus and Cryptococcus neoformans. PVAL at concentrations of 12.5 μg/mL was able to strongly inhibit Candida albicans and Staphylococcus aureus and 25 μg/mL Cryptococcus neoformans. The effect of PVAL on blood clotting was evaluated through activated partial thromboplastin time (APTT), prothrombin time (PT). Where PVLA promoted expressive prolongation of APTT, while for TP it promoted strong inhibition. In view of the above data, we can highlight that GaBL has high antitumor potential against cancer of the upper aerodigestive tract and has a notorious homeostatic activity. GaSL and PVAL have remarkable antifungal activity and can be a promising biotechnological tool, the latter having an important role in the homeostatic processes of coagulation, being a strong protractor of APTT and TP inhibitor. Finally, we emphasize that all the lectins studied have structural characteristics that favor the biotechnological study involving human health.

O crescimento da população mundial aumentou o investimento na agroindústria e
consequentemente aumentou a geração de resíduos agroindustriais sua maioria
lignocelulótico. Alternativas limpas de aproveitamento dos resíduos agroindustriais
vêm sendo pesquisadas em todo mundo, entre elas está à utilização dos resíduos
como fonte para crescimento fúngico e produção de enzimas de interesse
biotecnológico. Os Objetivos deste trabalho foram produzir, isolar, caracterizar e
aplicar biotecnologicamente a endoglucanase produzida a partir do fungo
filamentoso Pycnoporus sanguineus. Foram utilizados 5 diferentes resíduos
agroindustriais (farelo de trigo, bagaço de cana, pó de madeira, fibra de coco e papel
de filtro) por fermentação em estado sólido (FES). A enzima foi isolada através do
fracionamento etanólico e cromatografia de troca iônica, o isolamento foi confirmado
por (SDS-PAGE) em condição redutora e desnaturante. A endoglucanase isolada foi
caracterizada quanto a temperatura, pH, halotolerância, cinética enzimática e
aplicada biotecnologicamente através da sacarificação de resíduos agroindustriais. A
produção de endoglucanase foi maior no farelo de trigo (72 hs). A fração 80-100%
do fracionamento etanólico concentrou maior atividade enzimática e foi submetido a
cromatografia de troca iônica (DEAE-Sepharose), a enzima isolada foi eluída em
uma única fração, como confirmado no SDS-PAGE. A temperatura ótima da
endoglucanase isolada foi de 50°C e a estabilidade térmica entre 20° a 60°C com
ate 50% da atividade enzimática, temperaturas compatíveis com de outras
endoglucanases fúngicas. O pH ótimo foi o pH 5,0, com estabilidade em pH entre o
pH 4,5 e 8,0, demonstrando uma estabilidade em pH levemente alcalino compatível
com aplicações biotecnológicas em indústria de detergentes. A enzima isolada
manteve-se viável com atividade superior a 100% em até 5 M de NaCl, característica
importante pois alguns processos industriais acontecem na presença de grandes
quantidades de NaCl. A enzima apresentou Km 3,18 ± 100 mg/mL e o Kcat igual a
4,53 S-1 e conseguiu produzir açúcares redutores 273 mg/mL em 24 horas em casca
de arroz, mostrando-se viável para produção etanol de segunda geração. O
presente trabalho concluiu que o fungo P. sanguineus é produtor de uma
endoglucanase com caraterísticas bioquímicas importantes, de rápida produção e de
baixo custo.

The growth of the world population increased investment in agribusiness and,
consequently, increased the generation of agroindustrial residues, mostly
lignocellulosic. Clean alternatives for the use of agro-industrial residues have been
researched around the world, among them is the use of residues as a source for
fungal growth for the production of enzymes of biotechnological interest. The
objectives of this work were to produce, isolate, characterize and biotechnologically
apply the endoglucanase produced from the filamentous fungus Pycnoporus
sanguineus. Five different agro-industrial residues (wheat bran, sugarcane bagasse,
wood powder, coconut fiber and filter paper) were used by solid state fermentation
(FES). The enzyme was isolated through ethanol fractionation and ion exchange
chromatography, the isolation was confirmed by (SDS-PAGE) in reducing and
denaturing conditions. The isolated endoglucanase was characterized for
temperature, pH, halotolerance, enzymatic kinetics and applied biotechnologically
through the saccharification of agro-industrial residues. Endoglucanase production
was higher in wheat bran (72 hs). The 80-100% fraction of the ethanol fractionation
concentrated greater enzymatic activity and was subjected to ion exchange
chromatography (DEAE-Sepharose), the isolated enzyme was eluted in a single
fraction, as confirmed by SDS-PAGE. The optimal temperature of the isolated
endoglucanase was 50°C and the thermal stability was between 20° and 60°C with
up to 50% of the enzymatic activity, temperatures compatible with other fungal
endoglucanases. The optimum pH was pH 5.0, with stability at pH between pH 4.5
and 8.0, demonstrating stability at slightly alkaline pH compatible with
biotechnological applications in the detergent industry. The isolated enzyme
remained viable with activity greater than 100% in up to 5 M NaCl, an important
characteristic because some industrial processes take place in the presence of large
amounts of NaCl. The enzyme presented Km 3.18 ± 100 mg/mL and the Kcat equal
to 4.53 S-1 and managed to produce 273 mg/mL reducing sugars in 24 hours in rice
husks, proving to be viable for the production of second-generation ethanol . The
present work concluded that the fungus P. sanguineus is a producer of an
endoglucanase with important biochemical characteristics, fast production and low
cost.
